You have your files, and now you need to find material to file in them. Where are you going to get the ideas you need? Let's look at the speech material you might use category by category.
Anecdotes
The main source of anecdotes is surely your own memory. If you are speaking at a wedding reception, you must have known the groom for some time. There must be some incident in or aspect of his life that would make an interesting or amusing story. If you can't think of anything, perhaps photos in a photograph album will jog your memory.
If that doesn't help, ask other members of the family or friends of the groom if they can remember any interesting or amusing incidents. Someone is bound to have a tale to tell.
You could talk about how you met the groom, how you came to be friends, how the groom behaved differently after he had met the bride, the moment you knew it was serious, etc. There could be some behind-the-scenes stories about the wedding preparations or a story about something that happened during the bachelor party (though some of the goings-on at the bachelor party are better not divulged).
